Custom Jewelry Startup Gemvara Raises $25M In Oversubscribed Series D Round

gemvara_homepageGemvara, a Boston-based custom jewelry site we've written about in the past, just announced that it has closed a $25 million Series D funding round. This oversubscribed round was led by Norwest Venture Partners and also includes funds from all existing investors, including Highland Capital Partners, Canaan Partners and Balderton Capital. The company says that it plans to use this new influx of cash to "enhance all aspects of the customer shopping experience and continue to drive innovation that will bring the jewelry industry to the cutting edge of commerce." Gemvara previously raised a total of $20.2 million, including a large $15 million Series C round led by Balderton Capital.

Nintendo shows off new GamePad, new features

10 hrs.

In a video announcement featuring its president, Satoru Iwata, Nintendo showed off the final version of their new Wii U console and controller, along with some new social features like video chat and game-specific?message boards. The launch games will be shown on Tuesday at their on-stage presentation shortly before the doors open here at E3, but it seems Nintendo wanted to let a few cats out of the bag early.

The "new controller," as the company has been calling it publicly for a full year now, has finally been named: it's the Wii U GamePad. That's meant to be a throwback to the original NES gamepad as well as a fairly accurate description of the device itself: it's a pad that you game with.

The GamePad is similar to the prototype they showed off last year, but the layout is different, the whole controller is slightly?larger, and the circle pads in the top corners have been replaced with traditional analog sticks. There's also a spot for near-field communication (NFC), allowing tokens and cards to be scanned into the controller without swiping or putting in codes.

After Iwata explained the new GamePad (and quickly showed a sleek?new Pro controller, left) for those who don't want to use it), he moved onto Miiverse, Nintendo's new Mii-based platform for social interaction around games. Each game will have a community of players who can leave messages and tips inside the game --?several message?bubbles popped up?on one?Mario's defeat, for instance, with real people expressing frustration with that section and offering support.

Iwata said they feel this adds "a new degree of empathy between players," and also that "even with no one else in the room, you won't feel alone."?To that end, video communication between Wii U consoles will also be possible, using the camera built into the GamePad. And hand-written messages and doodles can be sent, a la Pictochat. Parts of this whole communication platform will also be available through a smartphone app.

Ways in which the Wii U will interact with the TV were teased but not fully fleshed out: the GamePad will act as a TV controller, for instance, and can be used to browse the internet or push video to the screen.

The actual games you'll be playing on the Wii U will be announced and shown off Tuesday morning at 9AM Pacific time; we'll be reporting live from the press conference, so watch this space for more news from Nintendo and the other majors here at E3.

More advanced therapies are being aimed at cancer

(AP) ? New research shows a sharp escalation in the weapons race against cancer, with several high-tech approaches long dreamed of but not possible or successful until now.

At a weekend conference of more than 30,000 cancer specialists, scientists reported:

?New "smart" drugs that deliver powerful poisons directly to cancer cells while leaving healthy ones alone.

?A new tool that helps the immune system attack a broad range of cancer types.

?Treatments aimed at new genes and cancer pathways, plus better tests to predict which patients will benefit from them.

"I see major advances being made in big diseases" such as breast and prostate cancers, said Dr. Richard Pazdur, cancer drug chief at the federal Food and Drug Administration, which on Wednesday announced a new policy intended to speed breast cancer drugs to the market.

The field continues to move toward more precise treatments with fewer side effects and away from old-style chemotherapy that was "like dropping a bomb on the body," he said.

In fact, an emerging class of "smart bombs" was one of the most hopeful developments reported at the meeting of the American Society of Clinical Oncology.

These are two-punch weapons that combine substances called antibodies, which bond with specific cancer cells, and toxins that are too potent to be given by themselves. A chemical link holds them together until they attach to a tumor cell, releasing the poison inside it and killing the cell.

"This is a classic example of the magic bullet concept" first proposed more than 100 years ago but only now possible with advances in technology, said Dr. Louis Weiner, director of Georgetown Lombardi Comprehensive Cancer Center.

"The antibody basically targets this very toxic drug right to the cancer cell and places it inside the cancer cell where the drug can do its damage" without harming healthy cells nearby, he said.

On Sunday, a large study showed that one such drug ? Genentech's T-DM1 ? delayed the time until cancer got worse in women with very advanced breast cancer. The drug also seems to be improving survival, although it will take more time to know for sure. So far, women on the new treatment were living more than a year longer than a comparison group of women who were given two other drugs.

Dozens of similar "smart bomb" drugs are in development. On Monday, Pfizer Inc. plans to report on one it is testing for certain types of lymphoma and leukemia. Only one such drug is on the market now ? Adcetris, sold by Seattle Genetics Inc. for some less common types of lymphoma.

The other big news at the conference involved a very different approach: using the immune system to fight cancer. For more than a century, doctors have been trying to harness its power, but tumor cells have cloaking mechanisms that have kept the immune system from recognizing them as "enemy" and going on the attack.

Bristol-Myers Squibb Co. has developed two drugs ? one aimed at cancer cells and the other at key soldier cells of the immune system ? to remove one of these invisibility cloaks. Two studies involving nearly 500 people found some tumor shrinkage in up to one quarter of patients with lung and kidney cancers as well as the deadly skin cancer, melanoma. The treatments had less impact against colon and prostate cancer.

These are only early results ? not survival comparisons or definitive tests, doctors warn. More testing is needed to even establish safety. In one study, three patients died of a lung inflammation considered due to the treatment.

However, ordinary chemotherapy can prove fatal, too, said one study leader, Dr. Julie Brahmer of Johns Hopkins University.

"There were a few patients who had a complete remission" from the immune system treatments and most patients suffered few side effects, she said. "It's great to see patients feeling well. They don't have hair loss, they don't have a drop in blood counts and are not as prone to infections."

Dr. Roy Herbst, medical oncology chief at Yale Cancer Center in New Haven, Conn., was hopeful.

"I haven't seen anything this good" for many years for treating lung cancer, he said. "I'd be very surprised if there wasn't some benefit" on survival, said Herbst, who has consulted for the drug's maker.

Other doctors, including Pfizer's cancer drug development chief, Dr. Mace Rothenberg, noted progress on new diagnostic tests to predict which drugs will work for which patients. Cost, time and difficulty have kept many of them from being practical in everyday settings for cancer patients, but "a lot of these barriers are falling," Rothenberg said.

"Every time we say 'this technology is 5 to 10 years off, we've been wrong" and progress has come sooner, he said.

LIBERTARIAN INVESTMENTS: Speculating and Investing

Today, I am going to talk about myself a bit and my experience in investing. ?I have been doing at least a little bit of investing for 15 years. ?It has consisted of stocks, mutual funds, commodities, options, money market funds, and probably other things that I can't think of right now.

When I look back at the big picture, there are really only two investment strategies that have paid off. ?One is in the permanent portfolio (mutual fund or otherwise) that was advocated in Harry Browne's book Fail Safe Investing. ?The other investment that has done well is commodities. ?This would be mostly gold and silver related investments, but also a little bit in energy.

When it comes to options or just trading individual stocks, I can't really say that I've done that well. ?I've certainly had some winners, but I've also had some losers. ?When I consider that these things almost offset each other over time, I probably would have been better off paying down my mortgage or putting more into the permanent portfolio setup.

I am guessing that I am not alone in this analysis. ?There are very few people who make it big in investing. ?Even most people who spend a significant amount of time studying the investment markets do not make out big, unless they are trading other people's money for a fee. ?In that case, it is really their job or career.

The problem is that many people feel they need to spend a lot of time doing their homework and watching CNBC for the latest stock tips. ?If they only understood the monetary system and understood the power of the permanent portfolio, it would save them a lot of time and headache. ?They could spend just a few minutes setting up their portfolio and then basically forget about it, except for rebalancing.

When it comes to speculating on stocks or any other investments, I am probably slightly better than average. ?Yet, I still acknowledge that it is usually a crap shoot and that it is impossible to time the markets. ?I cannot compete with Warren Buffett at his own game. ?Overall, playing the stock market is almost equivalent to playing blackjack in Las Vegas. ?In the case of the stock market, the big financial institutions and the government are the "house". ?They collect the most money in trading fees and taxes.

It can be fun to speculate in stocks, just like it is fun to play the lottery. ?But it really isn't that fun once you realize that you've lost. ?So my recommendation to most people is to save their money and be conservative with it. ?Pay down your debts. ?Invest in something similar to the permanent portfolio. ?Then take some money and pay down your mortgage. ?If you want to get ambitious, then buy an inexpensive investment property that will generate positive cash flow. ?After many years, you will be thankful that you were conservative with your money. ?You work hard for it, so you should watch over it carefully. ?If you want to speculate, then just set aside a small percentage of your savings as "play money".
